Pantoprazole belongs to the class of drugs called PROTON PUMP INHIBITORS which block the production of acid by the stomach. Other drugs in this group are Omeprazole, Rabeprazole. It acts by blocking the enzyme H+/ K+ ATPase better known as proton pump located in the secretary membrane of parietal cells. Whatever be the primary stimulus gastrin histamine or acetyl choline, it is the final activation of this pump which induces acid secretion. Thus, by blocking this pump it blocks the final common pathway for acid secretion

Pantoprazole is chemically more stable than omeprazole or lansoprazole in neutral or mildly acidic medium, but under strongly acidic conditions active species is formed.

INDICATIONS

Duodenal and gastric ulcer, Zollinger Ellison syndrome, gastro esophageal reflux disease.

 

CONTRA INDICATION

Hypersensitivity, pregnancy and lactation.

 

DRUG INTER ACTIONS

Less likely than omeprazole to react with other drugs. Pantoprazole reduces the absorption and concentration in blood of ketoconazole.

It increases the absorption and concentration in blood of digoxin. This may cause digoxin toxicity. Has low affinity for cytochrome P450

 

ADVERSE EFFECTS

Well tolerated. Diarrhea, dizziness, pruritis, skin rashes and gastrointestinal infections may occur rarely.
