Description
MECHANISM OF ACTION
Atorvastatin is a selective inhibitor of HMG CoA reductase the rate limiting enzyme that converts 3- hydroxy -3 -methyl glutaryl Co-enzyme a [HMG CoA] to mevalonate a precursor of sterols including cholesterol. Atorvastatin increases the level of HDL and decreases the level of LDL, VLDL and triglycerides. The mechanism of LDL lowering effect may involve both reduction of VLDL and induction of the LDL receptor, leading to reduced production or increased catabolism of LDL.
Atorvastatin decreases the LDL level in some patients with homozygous familial hyper cholesterolemia a population that rarely responds to other lipid lowering medication.
Atorvastatin reduces total cholesterol, LDL, and apoprotein B in patients with homozygous, heterozygous familial hypercholesterolemia, non-familial forms of hypercholesterolemia and dyslipidemia. Atorvastatin also reduces VLDL and triglycerides and produces variable increase in HDLand apolipoprotein A.
dose is recovered in urine.
